The rubber grips won't physically add a button to the vita, there might be a special case that does this but I haven't seen them before. The grips just make is easier for you to use the rear of the console without cramps.

If you press and hold the center of the screen on the Vita it shows you the control settings. Some games set R2 to be a corner of the touchscreen. Otherwise, i'd recommend buying a grip for the Vita which adds rubber handles to the side which makes it far easier to use the rear touch.
