Definitely. Especially if your using audio over a USB port, there's a good chance of lag. The only time it would matter is while in music production or while watching video. Have you watched a movie? Was the sound in sync?

I hate to say it, but this is going to be something that'll you'll have to deal with Samsung. Hopefully it's still under warranty. If you reset the TV and all of your other devices are still working, you have an internal problem with that TV. Check with Samsung or look for local TV repair...

In CCleaner, look under the Options tab for the Startup section. Remove anything that's not necessary for you in there. CCleaner's version is nice because it's tied in with the registry.
