-Take a screenshot of co-workers desktop
-Set as wallpaper
-Right click on desktop, go to "arrange icons by", then uncheck "show desktop icons". Also minimize the taskbar.
-Wait for co-worker to return
-Watch co-worker panic frantically as clicking does nothing.

Scheduling a task to run that locks their computer every minute works well, too I also like ejecting their CD tray remotely.
