How can I connect new Samsung flat screen tv audio outputs to my Bose 321 dvd player so that I can listen to tv through the Bose speakers (instead of soundbar). The Bose has yellow/white/red-spot jack plug inputs.

Connect the optical audio output of the TV to the optical audio input on the Bose. Check the TV manual on how to configure it. This will get you audio from the TV to the Bose.
You would also connect the Bose component or composite video out to the TV to get video from the Bose to the TV for DVD playback.
